Passage 1

Various methods are used to estimate the number of microorganisms in a culture. One method utilizes a spectrophotometer to measure the density of organisms in a broth medium. Density indicates the quantity of organisms in a given volume.

In this method, a beam of light is passed through the broth medium. The density of microorganisms in the broth is inversely proportional to the intensity of the light that passes through the broth. A meter is used to indicate the percentage of light transmitted through the medium.

The spectrophotometric data are used to produce a graph, such as the one below, that relates optical density to cell number.



Question: In Figure 1, if broth were added to the test tube in setup B, what would you expect the immediate result to be?


Choices:
A. Optical density would decrease.
B. Optical density would increase.
C. The growth of microorganisms would double.
D. The growth of microorganisms would decrease.



The correct answer is A.

The key word in the question is "immediate." The dilution of the broth would result in a decrease in optical density, as the same number of microorganisms would be in more broth. So (A) is correct.

(B) might have been tempting, as an increase in nutrients could eventually cause an increase in the growth of microorganisms. However, the immediate effect would be a decrease in optical density followed later by an increase in the growth of microorganisms.

Choice (C) is incorrect because there is no data to support any conclusion indicating by what factor growth might increase due to an increase in media.

Choice (D) is not correct because, in general, an increase in food supply promotes increases in the population of any living thing, not the reverse.
